Mechanism of Action
Huperzine A
Huperzine A is a potent, selective, and reversible inhibitor of acetylcholinesterase (AChE), binding to the enzyme's active site and preventing hydrolysis of acetylcholine to choline and acetate. By blocking AChE, it increases acetylcholine concentration in the synaptic cleft, prolonging activation of muscarinic (M1-M5) and nicotinic receptors. Huperzine A also blocks NMDA glutamate receptors in a non-competitive, use-dependent manner (similar to memantine), binding to the phencyclidine site within the ion channel and protecting neurons from excitotoxic calcium influx. It shows selectivity for the NR2A and NR2B subunits. Additionally, huperzine A has antioxidant properties, scavenging reactive oxygen species and reducing lipid peroxidation. It may enhance NGF signaling.
Uridine
Uridine (as UMP) is phosphorylated to UTP and enters the Kennedy pathway, where it combines with choline via CTP:phosphocholine cytidylyltransferase to form CDP-choline — the rate-limiting step in phosphatidylcholine synthesis. Uridine provides the nucleotide component needed for constructing phosphatidylcholine in neuronal cell membranes and synaptic vesicles. Uridine stimulates neurite outgrowth and synaptogenesis via activation of P2Y receptors and downstream PI3K/Akt signaling. It upregulates dopamine D2 receptor expression in the striatum and enhances dopaminergic neurotransmission. When combined with DHA (from fish oil) and choline, the three compounds synergistically increase synaptic membrane synthesis, dendritic spine density, and dopaminergic signaling — the 'Mr. Happy Stack' mechanism.
